About This Peptide
SS-31 (also known as Elamipretide or MTP-131) is a synthetic, mitochondria-targeted tetrapeptide (D-Arg-dimethylTyr-Lys-Phe-NH2). It is designed to freely cross cell membranes and selectively accumulate on the inner mitochondrial membrane. Rather than altering hormonal signaling or gene expression, SS-31 interacts directly with cardiolipin, a unique phospholipid crucial for maintaining inner mitochondrial membrane structure, to restore bioenergetics, boost ATP cellular energy, and inhibit mitochondrial oxidative stress.
Primary Actions in the Body
- Cardiolipin Stabilization & Structural Repair: Cardiolipin stabilizes the protein complexes of the electron transport chain (ETC) and curvature of mitochondrial cristae. Under oxidative stress or aging, cardiolipin oxidizes, causing membrane disruption. SS-31 binds selectively to cardiolipin, preventing its oxidation and preserving cristae structure.
- Optimization of ATP Synthesis: By restoring electron transport chain integrity (specifically Complex I and Complex IV), SS-31 optimizes the proton gradient across the inner membrane, significantly enhancing cellular ATP (energy) output in high-demand tissues like heart muscle, skeletal muscle, kidney, and brain.
- Targeted Reduction of Reactive Oxygen Species (mtROS): Uncoupled electron transport chains leak electrons that form harmful reactive oxygen species. By stabilizing the ETC complexes, SS-31 stops ROS generation at the source without acting as a non-specific systemic antioxidant that might disrupt normal cellular signaling.
- Inhibition of Ischemia-Reperfusion Injury & Apoptosis: When oxygen supply is cut off and restored (ischemia-reperfusion), mitochondria trigger cell death pathways. SS-31 prevents the opening of the mitochondrial permeability transition pore (mPTP) and blocks the release of cytochrome c, protecting cells from necrotic and apoptotic death.
